We have made a significant improvement to ToxTracker to increase the predictivity of the assay and to further expand the mechanisms of genotoxicity that are detected in the assay. ToxTracker consists of 6 different reporter cell lines for induction of DNA damage, oxidative stress and protein damage. The Bscl2-GFP and Rtkn-GFP genotoxicity reporters indicate the formation of DNA adducts and consequent inhibition of DNA replication, or the induction of DNA double strand breaks. The differential activation of these genotoxicity reporters allows for accurate identification of genotoxic compounds, discrimination between direct DNA reactivity and indirect genotoxicity and classification of compounds as clastogen or aneugen. We now enhanced the ToxTracker assay to directly discriminate clastogens from aneugens (i.e. tubulin poisons and kinase inhibitors). This extended version of the assay is named ToxTracker ACE (Aneugen Clastogen Evaluation). By including a DNA staining in the reporter cell lines during the ToxTracker analysis, we can directly assess the cell cycle and polyploidy status of cells following exposure. The combination of the differential activation of the ToxTracker genotoxicity reporters and polyploidy/cell cycle information is a unique and very powerful means to separate clastogens from aneugens and to identify tubulin poisons and kinase inhibitors. Below you can find an example of how this is applied to separate the clastogen Cisplatin from the tubulin poison Taxol and kinase inhibitor AMG900.

These platforms are used worldwide by large and small companies to source their testing services. Organisations listed as buyers can now make use of the standardised legal agreements of these platforms and efficiently order ToxTracker services.
ToxTracker Service Plan A prepaid ToxTracker service plan for easy testing at lower cost with a lower administrative burden. Service plans are available in packages of 25, 50 or 100 tests. Once a package is purchased, tests can be ordered in a series of projects within the duration of a year. ToxTracker results are delivered according to our usual turnaround time of 2 to 3 weeks.
