NAMs to the rescue: developmental toxicity of PROTACs
We evaluated the effects of seven drugs targeting different E3-ligases, including CRBN, VHL and MDM2, using the ReproTracker assay. All tested compounds were identified as teratogens.

Can ToxProfiler guide DILI predictions?
200 compounds with known toxicity profiles (64 for DILI) were screened in ToxProfiler. ToxProfiler correctly classified compounds with “most DILI concern” liabilities with high sensitivity (97%) and specificity (86%).

We made comparisons between ReproTracker and three in vitro teratogenicity high throughput assays, TeraTox, DevTox GLR, and hPST that have shorter assay durations.

Can ToxTracker replace the in vitro micronucleus test?
We tested 68 chemicals in ToxTracker and compared the results to in vitro and in vivo micronucleus and chromosome aberration assay data. ToxTracker showed a superior accuracy to correctly predict in vivo genotoxicity.
