ReproTracker 2.0: Pioneering Animal-Free Developmental Toxicity Testing with Enhanced Throughput and Efficiency

27th of August 2024 – Today, Toxys announces the higher throughput format of the ReproTracker assay for animal-free developmental toxicity predictions. ReproTracker 2.0 represents a novel advancement in the field of developmental toxicity testing that combines higher throughput, reduced compound requirement, and faster results, all while maintaining same sensitivity and specificity.

The miniaturized format allows for the use of robotic platforms, reducing the volume of compounds required and enabling high-throughput screening of multiple samples simultaneously. Moreover, during miniaturization process, differentiation protocols were optimized to reduce the assay time. Reduction in the assay’s duration not only lowers costs but also minimizes biological waste. The faster turnaround time accelerates the testing process, making it more feasible for early-stage screening.

ReproTracker 2.0 represents a significant step forward in the quest for animal-free testing methodologies, with the potential to reshape the landscape of toxicological assessment in the pharmaceutical, cosmetics, and chemical industries.

About ReproTracker 2.0

ReproTracker is a human induced pluripotent stem cells (hiPSCs)-based assay, which combines the assessment of functional/morphological changes with expression patterns of tissue-specific biomarkers to detect potential developmental toxicity responses of chemicals. The assay captures the key cellular events of stem cell differentiation and early embryonic development. ReproTracker is used to investigate if compounds interfere with early embryonic development and quantitatively predict their teratogenic potential.

In the validation study, the ReproTracker 2.0 maintained sensitivity and specificity comparable to previously developed ReproTracker. The assay can be used as an early-phase developmental toxicity screening platform, an alternative for animal-free teratogenicity testing of materials, or as a tool to extrapolate animal-derived results to humans.

Service information

Toxys offers ReproTracker as a service from our state-of-the-art laboratory in Leiden, the Netherlands. You can send your compounds (5-15 mg is required) and receive a full report within 6-8 weeks. The project report contains the protocol, technical details, concentration range finding results, biomarker expression with test compounds, morphological analysis with test compounds, ranking, and LOAEL concentrations of the test compounds. Please contact us with any questions you might have on ReproTracker.
