ReproTracker is a human induced pluripotent stem cell (hiPSCs)-based assay, which combines the assessment of changes in cell functionality/morphology and in expression patterns of tissue-specific biomarkers to detect developmental toxicity of novel drugs and chemicals.
During the webinar we will introduce the miniaturized version of the ReproTracker and its advantages when compared to its original format. The optimised assay presents key improvements, allowing for higher throughput, lower compound requirements, and faster results, all while maintaining similar sensitivity and specificity. The new miniaturized format is compatible with the use of automation and robotic platforms, majorly reduces the volume of compound required and enables high-throughput screening of multiple samples simultaneously. Moreover, this optimization process was characterised by the improvement of the differentiation protocols within ReproTracker, reducing not only the assay time but the overall span of the project.
Key improvements:
- Downscaled format: Shift from 24-well plates to 96-well plates.
- Increased throughput: 6-10 compounds per plate, compared to 4 compounds.
- Reduced turnaround time: Faster results in 6-8 weeks, down from 8-12 weeks.
- Lower compound requirement: Reduced from 100 mg to just 5-20 mg per test.
- Maintained performance: Sensitivity and specificity remain consistent, using the same biomarkers.
- Reduced pricing
ReproTracker 2.0 represents a significant step forward in the quest for animal-free testing methodologies, with the potential to reshape the landscape of toxicological assessment in the pharmaceutical, cosmetics, and chemical industries.
