Genotoxicity testing is an essential part of chemical risk assessment and includes testing for gene mutations, chromosomal aberrations, and numerical chromosomal changes. Traditionally, mutagenicity is measured by counting surviving cell clones after selection for gain/loss of function at a specific genetic locus. With the introduction of error-corrected sequencing, it is now possible to resolve precise MFs without selection at ultra-low frequencies, as low as 1×10-7.
MutaTracker is an all-in-one mammalian cell-based assay that combines mutation detection using SMM-seq with genotoxicity prediction using ToxTracker. SMM-seq, performed by Mutagentech, is a highly sensitive technique for detecting single nucleotide variants utilizing rolling circle amplification. This technique enables the comparison of several copies of the linearly amplified DNA to filter out artifacts and accurately identify true mutations. ToxTracker evaluates the induction of pathway-specific GFP-reporter genes to distinguish between primary genotoxicity and secondary genotoxicity caused by oxidative stress or protein damage.
