ToxTracker ACE: direct detection of clastogens and aneugens

We now enhanced the ToxTracker assay to discriminate clastogens from aneugens, and to classify aneugens as tubulin poisons or cell cycle kinase inhibitors. This extended version of the assay is named ToxTracker ACE (Aneugen Clastogen Evaluation).

ToxTracker consists of 6 different reporter cell lines to detect induction of DNA damage, oxidative stress and protein damage. The Bscl2-GFP and Rtkn-GFP genotoxicity reporters indicate formation of DNA adducts and consequent inhibition of DNA replication, or induction of DNA double strand breaks. Differential activation of these genotoxicity reporters allows for accurate identification of genotoxic compounds, discrimination between direct DNA reactivity and indirect genotoxicity and classification of compounds as clastogen or aneugen. By including a DNA staining in the reporter cell lines during the ToxTracker analysis, we can directly assess the cell cycle and polyploidy status of cells following exposure. 

The combination of the differential activation of the ToxTracker genotoxicity reporters and polyploidy/cell cycle information is a unique and very powerful means to separate clastogens from aneugens and to identify tubulin poisons and kinase inhibitors.
